| I. Background | |
| IL-17 家族是一個(gè)包含 6 種細(xì)胞因子的家族，包括 IL-17A、IL-17B、IL-17C、IL-17D、 IL-17E（也稱 IL-25）和 IL-17F。其中 IL-17A 是促炎信號(hào)的關(guān)鍵驅(qū)動(dòng)因子，由輔助性 T17 （Th17）細(xì)胞分泌，在自身免疫疾病中起著重要作用。該家族細(xì)胞因子在免疫反應(yīng)和炎癥 中發(fā)揮作用，并可能通過(guò)激活多種通路來(lái)介導(dǎo)信號(hào)轉(zhuǎn)導(dǎo)。IL17 受體家族包含 5 個(gè)成員（IL- 17RA 至 RE），其中 IL-17RA 是通用亞基，與配體特異性亞基（IL-17RB 結(jié)合 IL-25、IL- 17RC 結(jié)合 IL-17A/F、IL-17RE 結(jié)合 IL-17C）形成異源二聚體復(fù)合物。 配體結(jié)合后，受體通過(guò)胞內(nèi)保守的 SEFIR 基序招募銜接蛋白 ACT1，激活 TRAF6/TAK1 依賴性信號(hào)級(jí)聯(lián)，驅(qū)動(dòng) NF-κB 和 MAPK 通路活化，誘導(dǎo)促炎因子、抗菌肽 及基質(zhì)金屬蛋白酶（MMPs）表達(dá)。其過(guò)度激活直接導(dǎo)致銀屑病、哮喘等病理過(guò)程。 |
